AB OBJECTIVE: To determine the properties of Viruses produced by an SIVsm
chronically infected cell line. METHODS: Particles were examined by
e.m., by infectivity assays (in vitro and in vivo) and by immunoblots.
Nucleic acid was partially sequenced. RESULTS: Defective particles were
immature. The contained env and gag proteins but lacked vpx and vpr. A
1.6 kb genome deletion was observed. Particles were not infectious yet
they were immunogenic, and elicited neutralizing antibodies when
inoculated with and adjuvant to naire rhesus macaques. DISCUSSION AND
CONCLUSION: The approximately 1.6 kb deletion possibly involves
integrase, vif, vpr, and vpx. Some of these enzymes are required for
viral replication in vitro as well as in vivo. This immunogenic
particles could be used as a model for the study of vaccines against
pathogenic SIV.
